Rat Claudin-5/CLDN5 ELISA Kit

From Assay Genie, this Sandwich ELISA kit is designed for the quantitative measurement of claudin-5 (CLDN5) in rat samples. Claudin-5 is a critical component of tight junctions in endothelial cells, where it helps maintain blood-brain barrier integrity and regulate paracellular permeability. Dysregulation of claudin-5 has been associated with neurological disorders including stroke, multiple sclerosis, and Alzheimer's disease, making the kit a useful tool for research into vascular physiology, blood-brain barrier function, and related pathologies.

It is validated for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Linearity was demonstrated at 1:2, 1:4, and 1:8 dilutions in serum, EDTA plasma, and heparin plasma, and recovery averaged 94% in serum, 93% in EDTA plasma, and 94% in heparin plasma. The kit is stored at 2–8°C for 12 months.

The workflow uses a biotin-labeled detection antibody and HRP-Streptavidin, with TMB as the substrate and absorbance read at 450 nm. Incubations are performed at 37°C: 90 minutes for antigen binding, 60 minutes for detection antibody binding, 30 minutes for HRP-Streptavidin binding, and 10–20 minutes for color development.
