Rat Chromogranin-A (Chga) ELISA Kit

Spike-and-recovery in serum (n=5) showed 80-102% recovery (average 91%), EDTA plasma (n=5) 81-100% (average 90%), and heparin plasma (n=5) 80-89% (average 84%). Linearity across serial dilutions from 1:2 to 1:16 ranged from 79% to 104% across the three matrices tested. The sandwich ELISA is read at 450nm. The protocol involves adding 100µL of standard or sample per well and incubating for 2 hours at 37°C, followed by 100µL of Detection Reagent A for 1 hour at 37°C and washing three times. Detection Reagent B (100µL) is then added for a further 1 hour at 37°C with five washes, before adding 90µL of Substrate Solution for 15-25 minutes at 37°C and stopping with 50µL of Stop Solution prior to reading.
