Rat Choline O-acetyltransferase/CHAT ELISA Kit

This Sandwich ELISA kit from Assay Genie is designed for the quantitative measurement of Choline O-acetyltransferase (CHAT) in rat samples. The assay uses a double-antibody sandwich format with colorimetric detection, and is suitable for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. CHAT is described as a key enzyme involved in the synthesis of the neurotransmitter acetylcholine, with a role in neurotransmission and cognitive function, and has been implicated in conditions such as Alzheimer's disease and other neurological disorders.

The kit is reported to have a sensitivity of 0.188 ng/mL and an assay range of 0.313-20 ng/mL. In the procedure, standards and samples are incubated on the pre-coated plate at 37°C for 90 minutes, followed by a biotin-labeled detection antibody (60 minutes) and HRP-Streptavidin (SABC, 30 minutes); TMB substrate is then added for 10–20 minutes in the dark, the reaction is stopped, and absorbance is read at 450 nm. Linearity at 1:2, 1:4 and 1:8 dilutions is given as 85-90%, 84-91% and 83-98% for serum (n=5), 95-103%, 82-93% and 85-96% for EDTA plasma (n=5), and 87-105%, 82-98% and 85-97% for heparin plasma (n=5). Recovery ranges are 87-96% (average 93%) for serum, 88-101% (average 93%) for EDTA plasma, and 89-98% (average 94%) for heparin plasma. The kit is intended for research use in the neuroscience field and is stored at 2-8°C for up to 12 months.
