Description

From Assay Genie, this sandwich ELISA kit is designed for the quantitative measurement of Calcitonin Gene-Related Peptide (CGRP) in rat samples. It uses a double antibody configuration with a colorimetric detection method, and absorbance is read at 450 nm after addition of the stop solution. CGRP is a neuropeptide involved in modulating pain perception, inflammation, and vasodilation, and is a research target in conditions such as migraine, neurogenic inflammation, and neuropathic pain.

The kit accepts serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. It reports a sensitivity of 9.375 pg/mL and a measurable range of 15.625-1000 pg/mL. Linearity assessed in serum, EDTA plasma, and heparin plasma at 1:2, 1:4, and 1:8 dilutions gave recoveries of approximately 80% to 104%. Spiking recovery in the same matrices averaged 96-100%. Unopened reagents are stored at 2-8°C for up to 12 months.

The assay workflow comprises plate and reagent equilibration, a 90-minute primary incubation at 37°C, biotinylated detection antibody binding for 60 minutes, HRP-streptavidin incubation for 30 minutes, and TMB substrate development in the dark for 10–20 minutes. Sample preparation notes are supplied for serum (clot then 1000 × g for 20 minutes), plasma (anticoagulant tubes, 1000 × g for 15 minutes at 2-8°C), tissue homogenates, cell culture supernatants (2500 rpm for 5 minutes), and cell lysates.
