Description

Assay Genie offers a Sandwich ELISA (double antibody) kit for the quantitative measurement of Calcitonin Gene-Related Peptide (CGRP) in rat samples. The colorimetric assay is designed for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, and the kit components should be stored at 2-8°C for up to 12 months.

According to manufacturer specifications, the assay has a sensitivity of 9.375 pg/mL and a working range of 15.625-1000 pg/mL. Reported linearity for serum (n=5) spans 88-105% at 1:2, 85-104% at 1:4 and 85-102% at 1:8; EDTA plasma (n=5) shows 86-101%, 85-98% and 84-96% at the same dilutions; heparin plasma (n=5) shows 81-95%, 89-97% and 85-99%. Spike-and-recovery values are reported as 85-104% (average 96%) for serum, 86-102% (average 96%) for EDTA plasma, and 86-105% (average 97%) for heparin plasma.

The protocol follows a six-step sandwich workflow: reagent and plate preparation, primary incubation at 37°C for 90 minutes, biotin-labeled detection antibody incubation at 37°C for 60 minutes, HRP-Streptavidin (SABC) incubation at 37°C for 30 minutes, TMB color development in the dark for 10-20 minutes, and a stop reaction read at 450 nm. CGRP is described by the manufacturer as a neuropeptide involved in pain transmission, inflammation, and vasodilation, with elevated levels implicated in conditions such as migraines, arthritis, and neuropathic pain.
