Rat CDKN1A ELISA Kit

A Sandwich ELISA kit from Assay Genie uses a double-antibody format with colorimetric detection to quantify CDKN1A (Cyclin Dependent Kinase Inhibitor 1A, also known as p21) in rat samples. Compatible matrices include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The manufacturer states a sensitivity of 46.875 pg/mL and a measuring range of 78.125–5000 pg/mL, with storage at 2–8 °C for up to 12 months.

Reported linearity for serum (n = 5) is 90–103% at 1:2, 87–100% at 1:4 and 81–100% at 1:8; for EDTA plasma (n = 5) 85–99%, 94–100% and 84–100%; and for heparin plasma (n = 5) 85–100%, 86–96% and 81–90%. Spike-and-recovery averages are 92% for serum, 92% for EDTA plasma and 97% for heparin plasma.

The workflow includes primary incubation at 37 °C for 90 minutes, biotin-labelled detection antibody binding for 60 minutes, HRP-streptavidin incubation for 30 minutes, TMB substrate development in the dark for 10–20 minutes and absorbance reading at 450 nm after stop solution. The manufacturer describes CDKN1A as a key regulator of cell cycle progression whose dysregulation has been implicated in cancer and neurological disorders.
