Description

A sandwich ELISA kit from Assay Genie designed for the quantitative detection of CD278 in rat samples. CD278, also known as Inducible T-cell COStimulator (ICOS), is a critical protein involved in regulating T-cell activation and differentiation, with relevance to research in immune responses, inflammation, and immunotherapy. The kit uses a double-antibody sandwich format with colorimetric detection.

The assay is validated for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Reported sensitivity is 46.875 pg/mL, with a standard range of 78.125–5000 pg/mL. Storage is at 2–8°C for 12 months. Validation data reports recovery ranges of 86–105% for serum (n = 5, average 96%), 93–100% for EDTA plasma (n = 5, average 95%), and 91–105% for heparin plasma (n = 5, average 97%).

The procedure begins with reagent and plate preparation, followed by primary incubation at 37°C for 90 minutes to allow antigen binding. A biotin-labeled detection antibody is then added and incubated at 37°C for 60 minutes, followed by HRP-Streptavidin (SABC) for 30 minutes at 37°C. TMB substrate is added and the plate is incubated in the dark for 10–20 minutes; the reaction is stopped and absorbance is read at 450 nm.
