Description

A colorimetric Sandwich ELISA (Double Antibody) kit from Assay Genie designed for the precise measurement of CBS (Cystathionine beta-synthase) levels in rat samples. The kit is reactive with rat and validated for serum, plasma, cell culture supernatant, cell or tissue lysates, and other liquid samples. CBS is an enzyme essential for the metabolism of sulfur-containing amino acids and plays a critical role in homocysteine metabolism; its dysregulation has been linked to cardiovascular diseases, neurodegenerative disorders, and certain types of cancer, making this a useful reagent for metabolism research.

Performance characteristics reported by the manufacturer include a sensitivity of 9.375 ng/mL and a standard curve range of 15.625-1000 ng/mL. Recovery averages are 92% for serum (range 86-103%), 93% for EDTA plasma (range 86-104%), and 96% for heparin plasma (range 91-105%). Linearity across 1:2, 1:4 and 1:8 dilutions is reported for serum (85-105%), EDTA plasma (80-100%), and heparin plasma (80-105%). The associated Uniprot reference is P32232, and storage is at 2-8°C for 12 months. Synonyms listed include Cystathionine beta-synthase, Beta-thionase, Serine sulfhydrase, and Hemoprotein H-450.

The assay follows a standard sandwich workflow: equilibrate reagents and TMB substrate to room temperature and load standards, samples and blanks on the pre-coated plate; incubate at 37°C for 90 minutes for antigen binding; add biotin-labeled detection antibody and incubate at 37°C for 60 minutes; add HRP-Streptavidin (SABC) and incubate for 30 minutes; develop with TMB substrate in the dark for 10-20 minutes; add stop solution and read absorbance at 450 nm. Sample preparation guidance covers serum (clot, centrifuge at 1000 × g for 20 minutes), plasma (anticoagulant tubes, centrifuge at 1000 × g for 15 minutes at 2-8°C), tissue homogenate (homogenize in PBS with protease inhibitors), cell culture supernatant (centrifuge at 2500 rpm for 5 minutes), and cell lysates.
