Description

From Assay Genie, this sandwich ELISA kit is designed for the precise measurement of Cathepsin D (CTSD) levels in rat samples, including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Cathepsin D is a lysosomal aspartic endopeptidase involved in cellular processes such as protein degradation and autophagy, and its dysregulation has been linked to cancer, Alzheimer's disease, and osteoporosis, making it a valuable biomarker for research into these conditions and potential therapeutic targets.

The assay uses a colorimetric, double-antibody sandwich ELISA format with a sensitivity of 9.375 pg/mL and a working range of 15.625-1000 pg/mL (UniProt reference P24268). Reported linearity across 1:2, 1:4 and 1:8 dilutions is 85-101%, 82-101% and 89-100% for serum (n=5); 84-93%, 82-99% and 84-98% for EDTA plasma (n=5); and 87-103%, 85-103% and 86-96% for heparin plasma (n=5). Recovery averages are 95% for serum, 93% for EDTA plasma and 96% for heparin plasma. Reagents are stored at 2-8°C for 12 months.

The protocol includes equilibrating reagents to room temperature, incubating standards, samples and blanks on the pre-coated plate at 37°C for 90 minutes, then adding a biotin-labeled detection antibody (60 minutes at 37°C) and HRP-Streptavidin SABC (30 minutes at 37°C). TMB substrate is developed in the dark for 10-20 minutes, the stop solution is added, and absorbance is read at 450 nm. Sample preparation guidance covers serum, plasma, tissue homogenate, cell culture supernatant and cell lysate workflows.
