Rat Caspase-9 ELISA Kit

A sandwich ELISA from Assay Genie for the quantitative measurement of caspase-9 in rat samples. Caspase-9 is a key enzyme in the apoptotic pathway that plays a central role in programmed cell death, and its dysregulation has been linked to cancer, neurodegenerative disorders and cardiovascular diseases, making it a valuable biomarker for apoptosis research and therapeutic-target investigation.

The double-antibody sandwich assay uses colorimetric detection and is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Sensitivity is 9.375 pg/mL with a standard range of 15.625–1000 pg/mL. Spiked-recovery averages are 98% in serum (n = 5; range 93–103%), 94% in EDTA plasma (n = 5; range 88–104%) and 99% in heparin plasma (n = 5; range 91–104%). Linearity across 1:2, 1:4 and 1:8 dilutions is 82–100% / 81–98% / 85–97% for serum, 81–95% / 83–101% / 82–90% for EDTA plasma and 80–99% / 84–93% / 80–100% for heparin plasma (n = 5 each). The workflow is: equilibrate reagents and TMB substrate to room temperature and load standards, samples and a zero control onto the pre-coated plate, then incubate at 37°C for 90 minutes; add biotinylated detection antibody and incubate at 37°C for 60 minutes; add HRP-Streptavidin (SABC) and incubate at 37°C for 30 minutes; develop with TMB substrate in the dark for 10–20 minutes; stop the reaction and read absorbance at 450 nm. The kit is stored at 2–8°C and is stable for 12 months under those conditions.
