Rat Caspase 6/CASP6 ELISA Kit

A sandwich ELISA from Assay Genie for the quantitative measurement of Caspase-6 (CASP6) in rat samples. The kit supports research into apoptosis, where caspase-6 is described as a key enzyme in programmed cell death and whose dysregulation has been implicated in neurodegenerative disorders and cancer [S1, S2]. It accepts serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, and is read by colorimetric detection [C0, S1, S2].

The assay delivers a sensitivity of 18.75 pg/mL and operates across a range of 31.25-2000 pg/mL [S1, S2]. It is stored at 2-8°C for 12 months [S1, S2]. Linearity has been validated in serum, EDTA plasma and heparin plasma at 1:2, 1:4 and 1:8 dilutions, and recovery ranges of 92-99% (serum), 91-101% (EDTA plasma) and 87-102% (heparin plasma) have been demonstrated [S1, S2].

The six-step protocol proceeds as follows: equilibrate reagents and TMB substrate, then set up standards, samples and controls on the pre-coated plate; incubate at 37°C for 90 minutes to allow antigen binding; add biotin-labelled detection antibody and incubate at 37°C for 60 minutes; add HRP-Streptavidin (SABC) and incubate at 37°C for 30 minutes; develop with TMB substrate in the dark for 10–20 minutes; stop the reaction and read absorbance at 450 nm [S1, S2].
