Rat Caspase-1 ELISA Kit

This Sandwich ELISA kit from Assay Genie is designed for the quantitative measurement of caspase-1 in rat samples, including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Caspase-1 is a protein involved in inflammatory responses and programmed cell death, and its dysregulation has been linked to autoimmune disorders, infectious diseases, and metabolic disorders, making this kit a useful tool for cell death research.

The assay uses a double antibody sandwich format with a sensitivity of 37.5 pg/mL and a range of 62.5-4000 pg/mL. Reported linearity for serum (n = 5) spans 80-96% at 1:2, 87-105% at 1:4, and 89-98% at 1:8; EDTA plasma (n = 5) ranges from 82-101%, 88-101%, and 88-98% at the same dilutions; and heparin plasma (n = 5) spans 83-100%, 86-99%, and 89-98%. Recovery averages are 93% for serum, 99% for EDTA plasma, and 92% for heparin plasma. The kit should be stored at 2-8°C for up to 12 months.

The protocol involves equilibrating reagents and TMB substrate to room temperature, loading standards, samples and controls onto the pre-coated plate, and incubating at 37°C for 90 minutes. A biotin-labeled detection antibody is added and incubated at 37°C for 60 minutes, followed by HRP-Streptavidin (SABC) for 30 minutes. TMB substrate is then incubated in the dark for 10–20 minutes, the reaction is stopped, and absorbance is read at 450 nm.
