Description

From Assay Genie, this sandwich ELISA is configured as a double-antibody immunoassay with colorimetric readout for the quantification of Cardiotrophin-1 (CT-1) in rat-derived samples. Cardiotrophin-1 is described as a cytokine involved in cardiac development, function and maintenance, with roles in promoting cardiac hypertrophy and regulating the growth and survival of cardiomyocytes; dysregulation of CT-1 levels has been associated with various cardiovascular disorders, making it a biomarker of interest for heart disease research.

The kit accepts serum, plasma, cell culture supernatant, cell or tissue lysates and other liquid samples. Reported sensitivity is 9.375 pg/mL and the working range is 15.625-1000 pg/mL. Spiked-recovery testing gave ranges of 87-104% (average 97%) in serum (n=5), 92-104% (average 98%) in EDTA plasma (n=5) and 87-103% (average 95%) in heparin plasma (n=5). Linearity at 1:2, 1:4 and 1:8 dilutions was reported as 85-94%, 86-103% and 86-102% for serum, 82-94%, 85-99% and 84-97% for EDTA plasma, and 87-92%, 81-92% and 84-93% for heparin plasma. Reagents are stored at 2-8°C for 12 months.

The workflow comprises reagent and plate preparation, a primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody binding at 37°C for 60 minutes, HRP-streptavidin binding at 37°C for 30 minutes, TMB colour development in the dark for 10-20 minutes, and a stop step with absorbance read immediately at 450 nm.
