Rat CAPN1 ELISA Kit

Assay Genie's Sandwich ELISA Kit for rat calpain 1 (CAPN1) is designed for the precise measurement of calpain 1 levels in rat samples, with a colorimetric detection method suitable for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples.

The assay delivers a sensitivity of 0.375 ng/mL with a measurable range of 0.625–40 ng/mL. Validation data shows strong linearity across dilutions: serum (n=5) at 1:2/1:4/1:8 yielded 90–105% / 92–101% / 85–101%, EDTA plasma (n=5) 82–94% / 83–95% / 86–93%, and heparin plasma (n=5) 83–99% / 83–100% / 82–100%. Recovery averaged 98% for serum (93–103%), 97% for EDTA plasma (88–105%), and 95% for heparin plasma (86–105%). All components are intended for storage at 2–8°C for 12 months.

The protocol involves equilibrating reagents and TMB substrate to room temperature, incubating the plate at 37°C for 90 minutes for primary antigen binding, adding biotin-labeled detection antibody for a 37°C / 60-minute incubation, then HRP-Streptavidin (SABC) for 30 minutes. Colour development with TMB substrate takes 10–20 minutes in the dark, after which stop solution is added and absorbance is read at 450 nm.
