Rat C3a/Complement 3a ELISA Kit

Designed for the quantitative detection of C3a in rat samples, this Sandwich ELISA from Assay Genie employs a double-antibody, colorimetric format. The assay is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, and supports research investigating complement-mediated inflammation, immune response, and host defence mechanisms. C3a is a fragment of complement component C3, and its dysregulation has been linked to autoimmune disorders, inflammatory conditions, and infectious diseases.

Sensitivity is reported at 0.375 ng/mL, with a working range of 0.625–40 ng/mL. Linearity has been demonstrated in serum, EDTA plasma, and heparin plasma at 1:2, 1:4, and 1:8 dilutions, with serum at 1:2 ranging from 89–105%. Recovery across the same three sample types spans 85–105%, with average recoveries of 91% for serum, 91% for EDTA plasma, and 96% for heparin plasma. Reagents are stored at 2–8°C and remain stable for 12 months.

The protocol includes reagent and plate preparation, a 90-minute primary incubation at 37°C, biotin-labelled detection antibody binding (60 minutes at 37°C), HRP-streptavidin binding (30 minutes at 37°C), TMB colour development in the dark for 10–20 minutes, and absorbance reading at 450 nm after the stop solution is added. The kit is supplied with a pre-coated dismountable microplate, lyophilized standard, 100X biotin-labelled antibody, 100X HRP-streptavidin conjugate, TMB substrate, sample and antibody dilution buffers, SABC dilution buffer, stop solution, 25X wash buffer, plate sealers, and a technical manual.
