Description

Designed for quantitative measurement of Bone Morphogenetic Protein 3 in rat samples, this Assay Genie ELISA Kit uses a sandwich double-antibody format with colorimetric detection. The kit accepts serum, plasma, cell culture supernatant, cell or tissue lysates, and other liquid samples. BMP-3 is a critical signaling protein involved in bone development and regeneration, cartilage formation, cell differentiation, and tissue repair, and its dysregulation has been implicated in skeletal disorders, making it a valuable biomarker for research in orthopedics, regenerative medicine, and skeletal biology.

The assay reports a sensitivity of 37.5 pg/mL and a standard range of 62.5–4000 pg/mL, with reagents stored at 2–8°C for 12 months. Linearity in serum at 1:2, 1:4, and 1:8 dilutions is 93–102%, 94–104%, and 88–104% respectively; in EDTA plasma 82–97%, 92–100%, and 82–100%; and in heparin plasma 84–98%, 81–98%, and 84–99%. Average recovery is 97% for serum, 97% for EDTA plasma, and 98% for heparin plasma. The protocol comprises reagent and plate preparation, a 90-minute primary incubation at 37°C, biotin-labelled detection antibody incubation for 60 minutes at 37°C, HRP-streptavidin binding for 30 minutes at 37°C, TMB colour development for 10–20 minutes in the dark, and absorbance reading at 450 nm after stop solution addition.

Sample preparation covers serum (clot blood, centrifuge at 1000 × g for 20 minutes), plasma (anticoagulant tubes, centrifuge at 1000 × g for 15 minutes at 2–8°C), tissue homogenates (PBS with protease inhibitors), cell culture supernatant (centrifuge at 2500 rpm for 5 minutes), and cell lysates (lysis buffer with protease inhibitors).
