Rat beta-hexosaminidase A/beta-Hex A ELISA Kit

A colorimetric Sandwich ELISA (Double Antibody) from Assay Genie designed for the quantitative measurement of beta-hexosaminidase A (Beta-Hex A) in rat samples. The kit is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Detection relies on absorbance reading at 450 nm following TMB substrate development, with a reported sensitivity of 0.188 ng/mL and an assay range of 0.313–20 ng/mL. Reagents are stable when stored at 2–8°C for 12 months.

Beta-hexosaminidase A is a key enzyme involved in lysosomal storage disorders and plays a critical role in the breakdown of certain complex carbohydrates; its dysregulation has been associated with various diseases, making it a valuable biomarker for related research. Validated linearity (n = 5) is reported for serum at 1:2, 1:4 and 1:8 dilutions as 94–104%, 89–105% and 86–104%; for EDTA plasma as 85–101%, 94–99% and 82–94%; and for heparin plasma as 81–93%, 81–96% and 80–95%. Spike-and-recovery performance (n = 5) is reported for serum (average 97%), EDTA plasma (average 90%) and heparin plasma (average 95%).

The standard protocol comprises reagent and plate preparation, a 37°C primary incubation for 90 minutes, biotin-labelled detection antibody incubation for 60 minutes, HRP-streptavidin (SABC) binding for 30 minutes, TMB colour development in the dark for 10–20 minutes, and stop reaction with absorbance measurement at 450 nm. Sample preparation guidance covers serum, plasma, tissue homogenate, cell culture supernatant and cell lysate. The kit is supplied with a pre-coated ELISA microplate, lyophilised standard, biotin-labelled antibody (100X), HRP-streptavidin conjugate (SABC, 100X), TMB substrate, sample dilution buffer, antibody dilution buffer, SABC dilution buffer, stop solution, 25X wash buffer, plate sealer and a technical manual.
