Description

A colorimetric sandwich ELISA kit from Assay Genie designed for the precise measurement of Beclin-1 levels in rat samples. The double-antibody sandwich format delivers a sensitivity of 46.875 pg/mL with a working range of 78.125–5000 pg/mL. Compatible sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, with kit components stored at 2–8°C for 12 months.

Beclin-1 (UniProt Q91XJ1; synonyms Becn1 and Coiled-coil myosin-like BCL2-interacting protein) is a key regulator of autophagy, a process critical for cellular homeostasis and cell survival. Dysregulation of Beclin-1 has been implicated in cancer, neurodegenerative disorders, and infectious diseases, positioning the kit as a useful tool for immunology research into autophagy-related pathways.

The workflow involves equilibrating reagents and TMB substrate to room temperature, primary incubation at 37°C for 90 minutes, biotin-labeled detection antibody binding for 60 minutes, HRP-Streptavidin (SABC) incubation for 30 minutes, TMB color development in the dark for 10–20 minutes, and absorbance reading at 450 nm after adding stop solution. Reported recovery in serum samples (n = 5) ranges from 89–105% with an average of 94%, and linearity in serum at 1:2, 1:4, and 1:8 dilutions is 86–104%, 87–104%, and 88–103% respectively.
