Rat Axin 2 ELISA Kit

Axin 2 (UniProt O70240; synonyms include AXIN2, Axis Inhibitor 2, AXIL, Conductin, Axil, Axin-like protein, and Axis inhibition protein 2) is a key component of the Wnt signaling pathway, which governs cell growth, development, and differentiation, and its dysregulation has been linked to cancer, osteoporosis, and Alzheimer's disease, making it a valuable biomarker for related research.

Performance is defined by a sensitivity of 0.188 ng/mL and a working range of 0.313-20 ng/mL. Linearity across 1:2, 1:4 and 1:8 dilutions spans 81-104% for serum, EDTA plasma and heparin plasma (n=5). Spike-and-recovery testing returns average values of 93% in serum, 99% in EDTA plasma, and 93% in heparin plasma, with recovery ranges of 87-102%, 92-102%, and 89-103%, respectively. Absorbance is read at 450 nm following a TMB-based colorimetric step.

The procedure involves equilibrating reagents and TMB substrate to room temperature, a 37°C primary incubation of 90 minutes to allow antigen binding, a 60-minute incubation with the biotin-labelled detection antibody, a 30-minute HRP-Streptavidin (SABC) binding step, 10-20 minutes of color development with TMB in the dark, and immediate absorbance measurement at 450 nm after stop solution.
