Rat ATXN1 (Ataxin 1) CLIA Kit (RTES00050)

From Assay Genie, this chemiluminescent immunoassay (CLIA) kit is intended for the in vitro quantitative determination of Rat ATXN1 (Ataxin 1) in serum, plasma and other biological fluids, and is reactive with rat samples. The assay follows a Sandwich CLIA principle: a microplate pre-coated with antibody specific to Rat ATXN1 captures the analyte, after which a biotinylated detection antibody and an Avidin–Horseradish Peroxidase (HRP) conjugate are added sequentially. After washing, a substrate solution is added and the Relative Light Unit (RLU) value, measured on a chemiluminescence reader, is proportional to ATXN1 concentration, which is calculated against the standard curve. The kit is supplied for research use only.

Performance figures quoted in the technical manual are a sensitivity of 37.50 pg/mL and a detection range of 62.50–4000 pg/mL. No significant cross-reactivity or interference between Rat ATXN1 and analogues has been observed. The assay uses 100 µL of standard or sample per well with incubation steps at 37 °C: 90 minutes for sample binding, 1 hour for the biotinylated detection antibody, 30 minutes for the HRP conjugate, and up to 5 minutes for the substrate, protected from light. Working solutions of the detection antibody and HRP conjugate are prepared by diluting the supplied 100× concentrates. Sample preparation guidance is provided for serum, EDTA- or heparin-treated plasma, cell lysates, tissue homogenates, and cell culture supernatant or other biological fluids.

Kit components include a pre-coated CLIA plate, reference standard, 100× concentrated biotinylated detection antibody, 100× concentrated HRP conjugate, reference standard and sample diluent, biotinylated detection antibody diluent, HRP conjugate diluent, 25× concentrated wash buffer, substrate reagent A, substrate reagent B, plate sealer, desiccant and a product description. According to the manual, an unopened kit can be stored at 4 °C for 1 month, with the CLIA plate stored at -20 °C for 6 months and the HRP conjugate at -20 °C protected from light for 6 months; the diluents and substrate reagents are stored at 4 °C for 6 months.
