Rat ATP citrate lyase/ACLY ELISA Kit

Assay Genie supplies a Sandwich ELISA (Double Antibody) for the quantitative detection of ATP citrate lyase (ACLY) in rat samples, including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The procedure uses a biotin-labelled detection antibody, HRP-Streptavidin (SABC) and a TMB substrate, with absorbance read at 450 nm.

The reported sensitivity is 0.375 ng/mL and the working range is 0.625-40 ng/mL. Kits should be stored at 2-8°C for 12 months. Linearity in serum (n=5) is 90-104% at 1:2, 92-100% at 1:4 and 80-99% at 1:8; EDTA plasma (n=5) shows 87-95%, 84-101% and 81-100%; heparin plasma (n=5) shows 85-102%, 86-101% and 88-99%. Recovery averages are 96% in serum (range 85-101%), 98% in EDTA plasma (range 88-103%) and 98% in heparin plasma (range 93-101%).

ATP citrate lyase is a key enzyme in the citrate cleavage pathway with a central role in fatty acid biosynthesis and energy metabolism. The assay protocol runs at 37°C: 90 minutes primary incubation, 60 minutes detection antibody binding and 30 minutes HRP-Streptavidin binding, followed by 10-20 minutes of TMB colour development in the dark, stop solution and immediate absorbance reading at 450 nm. Recommended sample preparation includes serum clotted then centrifuged at 1000 × g for 20 minutes, and plasma centrifuged at 1000 × g for 15 minutes at 2-8°C.
