Rat ATF4 (Activating transcription factor 4) ELISA Kit

This Sandwich ELISA, Double Antibody kit from Assay Genie is designed for the quantitative detection of ATF4 (Activating Transcription Factor 4) in rat samples. The assay uses a colorimetric detection method and is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. It delivers a sensitivity of 28.125 pg/mL and a measuring range of 46.875-3000 pg/mL. ATF4 is a transcription factor involved in regulating stress responses, protein synthesis, and metabolism, and its dysregulation has been linked to cancer, diabetes, and neurodegenerative disorders, making this kit relevant for a broad range of research applications.

Reported performance characteristics include linearity across diluted matrices. For serum (n = 5), ranges of 88-103% (1:2), 85-97% (1:4) and 81-99% (1:8) are observed; EDTA plasma (n = 5) gives 87-103%, 89-99% and 80-96% at the same dilutions; heparin plasma (n = 5) gives 85-104%, 89-102% and 82-100%. Spike-and-recovery testing yields average recoveries of 93% for serum, 94% for EDTA plasma and 92% for heparin plasma, with respective ranges of 88-100%, 90-101% and 85-103%. The kit is stored at 2-8°C and remains stable for 12 months under these conditions.

The assay workflow begins with reagent and plate preparation at room temperature, followed by loading standards, samples and controls onto the pre-coated plate and incubating at 37°C for 90 minutes to allow antigen binding. A biotin-labelled detection antibody is then added and incubated at 37°C for 60 minutes, after which HRP-Streptavidin conjugate (SABC) is incubated for 30 minutes. TMB substrate is added and incubated in the dark for 10-20 minutes, the reaction is stopped, and absorbance is read at 450 nm.
