The aryl hydrocarbon receptor is a key transcription factor involved in xenobiotic metabolism, immune response, and cell differentiation, with dysregulation linked to cancer, autoimmune disorders, and developmental abnormalities. Linearity was validated for serum, EDTA plasma, and heparin plasma across 1:2, 1:4, and 1:8 dilutions, with values ranging from 80-105% depending on sample and dilution. Recovery ranges from 85-105% across these sample types, with average recovery of 98% for serum, 94% for EDTA plasma, and 96% for heparin plasma.

The assay involves equilibrating reagents to room temperature, incubating standards and samples at 37°C for 90 minutes for antigen binding, adding biotin-labeled detection antibody for 60 minutes at 37°C, HRP-streptavidin for 30 minutes, TMB substrate for 10-20 minutes in the dark, and reading absorbance at 450 nm after adding stop solution.
