Description

This Sandwich ELISA kit from Assay Genie is designed for the precise measurement of Apolipoprotein B (APOB) levels in rat samples. Apolipoprotein B is a key protein involved in lipid transport and metabolism, playing a crucial role in the regulation of cholesterol levels and cardiovascular health. Abnormalities in APOB levels have been linked to conditions such as atherosclerosis, cardiovascular diseases, and metabolic disorders, making it a valuable biomarker for studying these diseases and potential therapeutic interventions.

The assay has a sensitivity of 1.875 ng/mL and a detection range of 3.125–200 ng/mL. Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit uses a colorimetric, double-antibody sandwich format, with standards, samples and controls loaded onto a pre-coated plate and absorbance read at 450 nm after TMB substrate development. Reagents are stored at 2–8 °C, with a shelf life of 12 months under appropriate storage conditions.

Recovery was evaluated in spiked matrices: serum (n=5) 86–98% (average 91%), EDTA plasma (n=5) 88–104% (average 98%), and heparin plasma (n=5) 85–105% (average 98%). Linearity was demonstrated across serial dilutions (1:2, 1:4, 1:8) for serum, EDTA plasma and heparin plasma, with results expressed as a percentage of the expected concentration. The assay procedure involves equilibrating reagents and TMB substrate to room temperature, primary incubation at 37 °C for 90 minutes, biotin-labelled detection antibody incubation for 60 minutes, HRP-streptavidin binding for 30 minutes, and colour development in the dark for 10–20 minutes before stopping and reading absorbance at 450 nm.
