Description

From Assay Genie, this Sandwich ELISA kit is designed for the quantitative detection of intestinal alkaline phosphatase (ALPI) in rat-derived samples, including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The double-antibody sandwich format is read colorimetrically, and ALPI is also known by the synonyms Alpi, IAP-I, and intestinal alkaline phosphatase I.

Performance characteristics reported by Assay Genie include a sensitivity of 0.469 ng/mL and a working range of 0.781–50 ng/mL. Linearity has been assessed in serum, EDTA plasma, and heparin plasma (n = 5) at 1:2, 1:4, and 1:8 dilutions, with recovery across the same sample types ranging from 86% to 104% depending on matrix and dilution. The kit is stored at 2–8°C for 12 months.

The protocol involves equilibrating reagents and TMB substrate to room temperature, loading standards, samples and blanks for a 90-minute primary incubation at 37°C, addition of a biotin-labelled detection antibody with a 60-minute incubation at 37°C, HRP-streptavidin binding for 30 minutes at 37°C, TMB colour development in the dark for 10–20 minutes, and absorbance measurement at 450 nm after adding stop solution. ALPI is an enzyme involved in intestinal health and function, with abnormal levels associated with gastrointestinal and inflammatory bowel disorders, making it a useful biomarker for gastrointestinal research.
