Rat alpha 2 Macroglobulin ELISA Kit

A sandwich ELISA from Assay Genie designed for the quantitative measurement of alpha-2-macroglobulin in rat samples. The kit uses a double-antibody sandwich format with colorimetric detection and is suitable for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Alpha-2-macroglobulin is relevant to research into inflammation and tissue repair, and its levels are altered in disease states including inflammatory conditions, cardiovascular disease, and neurodegenerative disorders.

Reported performance includes a sensitivity of 0.281 ng/mL and a standard range of 0.469–30 ng/mL. Linearity was demonstrated at 1:2, 1:4 and 1:8 dilutions in serum (88–101%, 82–98%, 83–94%), EDTA plasma (95–100%, 85–99%, 81–96%) and heparin plasma (99–105%, 82–101%, 86–100%). Recovery was 89–105% in serum (average 99%), 88–100% in EDTA plasma (average 93%) and 91–103% in heparin plasma (average 97%). The kit is stored at 2–8°C for up to 12 months.

The workflow comprises reagent and plate equilibration, loading of standards and samples onto the pre-coated plate with incubation at 37°C for 90 minutes, addition of a biotin-labeled detection antibody (37°C for 60 minutes), HRP-Streptavidin binding (37°C for 30 minutes), TMB substrate development in the dark for 10–20 minutes, and absorbance reading at 450 nm after addition of stop solution.
