Rat Agtr2/Type-2 angiotensin II receptor ELISA Kit

Assay Genie provides this Sandwich ELISA for the quantitative detection of AGTR2 (Type-2 angiotensin II receptor) in rat samples, using a double-antibody colorimetric method. AGTR2 is a key receptor in the angiotensin II signaling pathway, which regulates blood pressure, fluid balance, and vascular biology. Dysregulation of this pathway is implicated in cardiovascular diseases, kidney disorders, and neurological conditions.

The kit is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Sensitivity is 46.875 pg/mL, with a measuring range of 78.125–5000 pg/mL. Storage is 2–8°C for 12 months. Linearity (n=5) at 1:2, 1:4, and 1:8 dilutions was 94–103%, 87–97%, and 91–105% for serum; 83–98%, 82–101%, and 84–100% for EDTA plasma; and 91–100%, 82–99%, and 81–95% for heparin plasma. Recovery ranges with averages were 91–101% (97%) for serum, 88–97% (93%) for EDTA plasma, and 95–104% (99%) for heparin plasma.

The workflow includes equilibrating reagents and TMB substrate to room temperature, loading standards and samples onto the pre-coated plate, incubating at 37°C for 90 minutes for primary antigen binding, adding biotin-labeled detection antibody for 60 minutes, adding HRP-Streptavidin (SABC) for 30 minutes, developing with TMB substrate in the dark for 10–20 minutes, stopping the reaction, and reading absorbance at 450 nm. Sample preparation protocols are given for serum (allow to clot, centrifuge at 1000 × g for 20 minutes), plasma (anticoagulant tubes, centrifuge at 1000 × g for 15 minutes at 2–8°C), tissue homogenate, cell culture supernatant, and cell lysate.
