This competitive immunoassay from Assay Genie is built around a microtitre plate pre-coated with a rat adiponectin (ADPN) conjugate and is calibrated to quantify adiponectin, a 30 kDa adipokine hormone secreted primarily by white adipose tissue. Adiponectin plays a central role in regulating glucose homeostasis, fatty acid oxidation, and insulin sensitivity, and circulating concentrations are inversely correlated with body mass index in rodent models of diet-induced obesity and type 2 diabetes. Researchers commonly apply this kit to serum samples, plasma samples, and cell culture supernatant collected from Sprague-Dawley or Wistar rats to monitor metabolic changes following high-fat diet feeding, pharmacological intervention, or bariatric surgery protocols.

The assay uses a competitive binding principle in which endogenous adiponectin in the sample competes with a labelled reference antigen for binding sites on a specific antibody, and the resulting complex is visualised with an enzyme-substrate reaction quantified on a standard plate reader. Each kit supplies a pre-coated microplate, assay buffer, detection antibody, reference standards, wash concentrate, substrate solution, stop solution, and a sealed plate cover, enough reagents for a full plate of unknown samples run in duplicate alongside a seven-point standard curve. Unopened kits are dispatched on cold packs; for routine storage the sealed plate and working reagents should be refrigerated at 4 °C, while reconstituted standards should be frozen in single-use aliquots to limit repeated freeze-thaw cycles and preserve immunoreactivity.

Intended for research use only, this kit is not approved for diagnostic procedures in humans. Typical end-users include academic and pharmaceutical laboratories investigating metabolic syndrome, cardiovascular risk biomarkers, non-alcoholic fatty liver disease, and adipose tissue biology in rat models, where the competitive format is well matched to the relatively high endogenous circulating levels of adiponectin and avoids the dilution bias seen with two-antibody capture formats. Pairing the ELISA readout with western blot analysis of adipose lysates or PCR profiling of adiponectin receptor transcripts gives a more complete picture of pathway activity in vivo.
