The protocol follows a standard sandwich workflow: equilibrate reagents to room temperature, incubate the pre-coated plate at 37°C for 90 minutes for antigen binding, add biotin-labelled detection antibody and incubate for 60 minutes, incubate with HRP-Streptavidin (SABC) for 30 minutes, develop with TMB substrate in the dark for 10-20 minutes, and read absorbance at 450 nm immediately after adding stop solution.
