Rat ADAMTS4 ELISA Kit

From Assay Genie, this sandwich ELISA kit is designed for the quantitative measurement of ADAMTS4 in rat samples. ADAMTS4 is an enzyme involved in the breakdown of cartilage and extracellular matrix proteins, with relevance to research in osteoarthritis and inflammatory diseases. The kit uses a double antibody sandwich format with colorimetric detection, offering a sensitivity of 46.875 pg/mL and a working range of 78.125-5000 pg/mL.

Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Linearity for serum (n=5) ranges 89-103% at 1:2, 91-103% at 1:4, and 87-103% at 1:8 dilution. EDTA plasma (n=5) shows 82-97% at 1:2, 87-101% at 1:4, and 84-94% at 1:8. Heparin plasma (n=5) shows 83-100% at 1:2, 81-96% at 1:4, and 84-99% at 1:8. Recovery ranges are 90-105% for serum (average 98%), 85-100% for EDTA plasma (average 93%), and 86-93% for heparin plasma (average 90%). The assay is read at 450 nm following TMB substrate development, and components should be stored at 2-8°C for 12 months.

The protocol involves equilibrating reagents to room temperature, a 90-minute primary incubation at 37°C, biotin-labelled detection antibody binding for 60 minutes, HRP-streptavidin incubation for 30 minutes, and TMB colour development in the dark for 10-20 minutes before addition of stop solution and absorbance measurement.
