Description

This double-antibody sandwich ELISA from Assay Genie is designed for the quantitative detection of prostatic acid phosphatase (ACPP) in rat samples. The colorimetric assay is read at 450 nm and targets ACPP, an enzyme linked to phosphate metabolism and prostate cancer research.

The kit reports a sensitivity of 18.75 pg/mL and a working range of 31.25-2000 pg/mL, with reactivity for rat. Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Storage is at 2-8°C for 12 months. Linearity in serum (n=5) was 89-102% at 1:2, 85-94% at 1:4, and 91-99% at 1:8; EDTA plasma (n=5) gave 89-95%, 85-99%, and 91-100% at the same dilutions, while heparin plasma (n=5) gave 91-105%, 85-98%, and 91-97%. Recovery averaged 94% (range 91-97%) for serum, 96% (range 92-98%) for EDTA plasma, and 93% (range 86-99%) for heparin plasma.

The workflow begins with equilibrating reagents and TMB substrate to room temperature, followed by loading standards, samples, and a zero control onto the pre-coated plate for a 90-minute incubation at 37°C. A biotin-labelled detection antibody is then added for 60 minutes, followed by HRP-streptavidin (SABC) for 30 minutes. TMB substrate is added for 10-20 minutes in the dark, and absorbance is measured at 450 nm after adding the stop solution.
