Rat Acid Phosphatase/ACP ELISA Kit

Assay Genie provides a colorimetric Sandwich ELISA built on a double-antibody format for the quantitative measurement of Acid Phosphatase (ACP) in rat-derived samples. The kit is validated for use with serum, plasma, cell culture supernatants, cell or tissue lysates, and other liquid samples, supporting research into ACP biology and its reported roles in bone formation, immune system function, and metabolic regulation.

Reported sensitivity is 0.469 ng/mL with a working assay range of 0.781–50 ng/mL. Linearity in serum (n=5) spans 85–104% at 1:2, 92–97% at 1:4 and 89–105% at 1:8 dilutions; EDTA plasma gives 92–99%, 90–101% and 83–100%, and heparin plasma gives 83–96%, 83–94% and 84–100% across the same dilutions. Spike-and-recovery averages are 93% for serum, 91% for EDTA plasma and 96% for heparin plasma across n=5.

The six-step protocol is: equilibrate reagents and TMB substrate at room temperature, load standards and samples and incubate the plate at 37°C for 90 minutes, add biotin-labelled detection antibody and incubate at 37°C for 60 minutes, add HRP-Streptavidin (SABC) for 30 minutes at 37°C, develop with TMB substrate for 10–20 minutes in the dark, then add stop solution and read absorbance at 450 nm. Unopened kits should be stored at 2-8°C for 12 months.
