Rat ABCB1/P Glycoprotein ELISA Kit

From Assay Genie, this Sandwich ELISA, Double Antibody kit is specifically designed for the accurate and precise measurement of ABCB1 levels in rat samples including serum, plasma, cell culture supernatants, cell or tissue lysates, and other liquid samples. The assay uses a colorimetric detection method. ABCB1, also known as P-glycoprotein, is a key transporter protein involved in the efflux of various substances across cell membranes, and its role in drug resistance and impact on the pharmacokinetics of many drugs make it a crucial target for research in pharmacology and toxicology.

The kit has a sensitivity of 0.188 ng/mL and an assay range of 0.313-20 ng/mL. Linearity was validated in serum (94-105%, 85-101%, 85-105% at 1:2, 1:4 and 1:8 dilutions; n=5), EDTA plasma (89-100%, 92-101%, 88-101%; n=5) and heparin plasma (82-98%, 80-98%, 82-88%; n=5). Recovery ranges were 89-101% (average 94%) for serum, 86-103% (average 96%) for EDTA plasma, and 85-103% (average 94%) for heparin plasma (n=5). The kit should be stored at 2-8°C for 12 months.

The assay procedure follows six steps: reagent and plate preparation; primary incubation at 37°C for 90 minutes; biotin-labeled detection antibody binding at 37°C for 60 minutes; HRP-Streptavidin binding at 37°C for 30 minutes; TMB substrate color development in the dark for 10–20 minutes; and a stop reaction with absorbance read at 450 nm.
