Description

Designed for accurate measurement of RAD51 protein levels in cell lysates and tissue samples, this Cell-Based ELISA Kit from Assay Genie uses a colorimetric detection method measured at 450 nm and is reactive with Human and Mouse samples. The kit is supplied in a 96-Well Microplate format, has a dynamic range of > 5000 Cells, and offers storage stability of 4°C for 6 Months. It is suitable for adherent cells and is intended for research areas including Epigenetics and Nuclear Signaling.

The kit is a lysate-free, high-throughput and sensitive assay that can detect the RAD51 protein expression profile in cells. It is used to measure the relative amounts of RAD51 in cultured cells and to screen for the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on RAD51. Qualitative determination of RAD51 concentration is achieved by an indirect ELISA format: RAD51 is captured by RAD51-specific primary antibodies, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, allowing the conjugated HRP to catalyze a colorimetric reaction upon substrate addition.

Because the assay is qualitative in nature, two normalization methods are provided. A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control for normalizing target absorbance values. Following colorimetric measurement of HRP activity, a Crystal Violet whole-cell staining method may be used to determine cell density, with absorbance values then normalized to cell amounts to adjust for plating differences.
