Description

Assay Genie's colorimetric cell-based ELISA kit is designed for the qualitative measurement of RAD17 protein levels in cultured cells. The assay uses an indirect ELISA format in which the target is captured by RAD17-specific primary antibodies and detected using HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition.

The kit is described as lysate-free and suitable for high-throughput applications, enabling profiling of RAD17 expression in cells and screening the effects of treatments, inhibitors (such as siRNA or chemicals), or activators. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalisation, and Crystal Violet whole-cell staining may be used to determine cell density after the colorimetric readout. Detection is performed at 450 nm, the stated dynamic range is > 5000 Cells, and the kit is reactive with human and mouse samples. Storage is at 4°C with a stability of 6 months.

RAD17 is a cell cycle checkpoint protein involved in the DNA damage response, helping to coordinate cell cycle arrest and DNA repair. It shares strong similarity with DNA replication factor C (RFC), binds chromatin prior to DNA damage, and is phosphorylated by ATR following damage, inducing G2 arrest. The gene is catalogued under Gene ID 5884 and UniProt ID O75943, supporting applications in cell cycle, DNA damage, and oncology research.
