Rabphilin-3A is described as a vital protein involved in intracellular vesicle trafficking and neurotransmitter release, and its dysfunction has been implicated in various neurological disorders, making it a useful biomarker for studying these conditions and for developing therapeutic interventions. Qualitative determination of Rabphilin 3A concentration is achieved by an indirect ELISA format: target is captured by Rabphilin 3A-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining method is provided for determining cell density so absorbance values can be normalised to cell amounts.
