Rabbit MMP 9 ELISA Kit

From Assay Genie, this sandwich ELISA kit with a double-antibody configuration enables quantitative measurement of rabbit matrix metalloproteinase-9 (MMP-9) in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The colorimetric assay delivers results across a measuring range of 1.563–100 ng/mL with a sensitivity of 0.938 ng/mL, supporting research into MMP-9's role in extracellular matrix breakdown and tissue remodelling — processes relevant to wound healing, immune response and tumour invasion. Aberrant MMP-9 levels have been linked to cancer, arthritis and cardiovascular disorders, highlighting the analyte's value as a biomarker for disease progression and therapeutic development studies.

Spiked-recovery testing in serum gave an average of 98% (range 88–105%), in EDTA plasma an average of 93% (range 86–104%), and in heparin plasma an average of 94% (range 85–104%). Linearity was assessed at 1:2, 1:4 and 1:8 dilutions: serum returned 86–94%, 82–99% and 80–98%; EDTA plasma returned 87–104%, 82–97% and 84–100%; and heparin plasma returned 91–99%, 82–98% and 85–99%. The assay uses a pre-coated plate: reagents and TMB substrate are equilibrated to room temperature, then standards, samples and blanks are loaded and incubated at 37°C for 90 minutes. After washing, biotin-labelled detection antibody is added and incubated at 37°C for 60 minutes, followed by HRP-Streptavidin (SABC) for 30 minutes at 37°C. Colour is developed with TMB substrate for 10–20 minutes in the dark, then stop solution is added and absorbance is read at 450 nm.

Sample-preparation guidance is supplied in the manual: serum is allowed to clot then centrifuged at 1000 × g for 20 minutes; plasma is collected in anticoagulant tubes and centrifuged at 1000 × g for 15 minutes at 2–8°C. Kit reagents are stored at 2–8°C for 12 months, with the biotinylated antibody, SABC and TMB substrate additionally protected from direct light. The protocol included with the kit is specific to each batch/lot and should be followed in preference to any sample protocol.
