Description

From Assay Genie, this rabbit-reactive sandwich ELISA uses a double-antibody format with colorimetric detection to provide precise measurement of MCP-1, also known as CCL2. MCP-1 is a key chemokine involved in recruiting monocytes and macrophages to sites of inflammation and is implicated in conditions such as atherosclerosis, rheumatoid arthritis, and asthma, making the kit a useful tool for studies of inflammation and immune response.

The kit is intended for use with rabbit serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Sensitivity is 18.75 pg/mL with a measuring range of 31.25-2000 pg/mL. Reported recovery averages are 94% for serum (n = 5, range 85-103%), 94% for EDTA plasma (n = 5, range 86-105%), and 100% for heparin plasma (n = 5, range 90-104%). Linearity across 1:2, 1:4 and 1:8 dilutions ranges from 83% to 105% across serum, EDTA plasma and heparin plasma. Reagents are stored at 2-8°C for 12 months.

The protocol involves equilibrating reagents and TMB substrate to room temperature, loading standards and samples for a 90-minute primary incubation at 37°C, binding with a biotin-labeled detection antibody for 60 minutes, adding HRP-Streptavidin (SABC) for 30 minutes, developing with TMB substrate in the dark for 10-20 minutes, and reading absorbance at 450 nm after adding stop solution. Synonyms associated with the target include MCP-1, CCL2, GDCF-2, HC11, HSMCR30, MCAF, MCP1, SCYA2, and SMC-CF.
