Leptin (LEP) is a 16 kDa adipocyte-derived hormone that regulates appetite, energy expenditure, and body weight through hypothalamic signaling pathways. The Assay Genie Rabbit Leptin ELISA Kit is an in vitro quantitative immunoassay developed for research use only, calibrated to measure endogenous leptin in rabbit-derived specimens using a sequential antibody binding workflow read on a standard microplate spectrophotometer.

Standards and samples are incubated in pre-coated microplate wells, followed by a biotinylated detection antibody and a streptavidin-enzyme conjugate that produces a chromogenic response when developed with TMB substrate. The reaction is stopped with an acid solution, and absorbance values are interpolated against the supplied recombinant rabbit leptin calibration curve, allowing quantification across the dynamic range of the assay.

Research applications include metabolic and obesity studies, nutritional physiology investigations, characterization of rabbit models for diabetes and cardiovascular disease, reproductive endocrinology work, and pharmacologic screening of compounds that influence adipokine secretion. Because the calibrators are rabbit leptin, results from other species should not be inferred from this product, and the kit is intended only for laboratory research applications rather than diagnostic use.

The kit supplies a 96-assay microplate pre-coated with capture antibody, lyophilized recombinant rabbit leptin standards, biotinylated detection antibody, streptavidin-HRP conjugate, TMB substrate, wash buffer concentrate, stop solution, and assay diluent for 96 individual determinations. Unused strips should be resealed in the foil pouch with desiccant and refrigerated between sessions; reconstituted standards and detection antibody should be aliquoted into single-use vials to avoid repeated freeze-thaw cycles and stored at -20 °C per the supplied protocol.

For sample preparation, whole blood should be processed promptly and the resulting serum or plasma stored frozen at -80 °C in single-use aliquots. Tissue samples can be homogenized in PBS containing protease inhibitors and clarified by centrifugation at 5000 x g for 10 minutes before assay. Researchers are advised to test serial dilutions of each matrix because lipemic or hemolyzed specimens may require additional dilution to fall within the linear portion of the standard curve.
