Description

From Assay Genie, this Sandwich ELISA enables quantitative measurement of inducible nitric oxide synthase (iNOS) in rabbit samples, including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The double-antibody assay uses a colorimetric detection method in which TMB substrate is added and absorbance is read at 450 nm after a stop solution.

iNOS is an enzyme that plays a key role in the production of nitric oxide, a signalling molecule involved in immune response and inflammation, and dysregulation of its activity has been associated with inflammation, infection, and neurodegenerative disorders. The kit reports a sensitivity of 0.188 ng/mL and a measuring range of 0.313–20 ng/mL, with components intended for storage at 2-8°C for 12 months.

The protocol proceeds through reagent and plate preparation, primary incubation at 37°C for 90 minutes, biotinylated detection antibody binding for 60 minutes, HRP-streptavidin binding for 30 minutes, TMB colour development in the dark for 10–20 minutes, and final absorbance reading. Reported linearity across 1:2, 1:4 and 1:8 dilutions is 80–101% for serum, 82–105% for EDTA plasma, and 82–105% for heparin plasma, with average recoveries of 96% (serum), 93% (EDTA plasma) and 93% (heparin plasma).
