This sandwich ELISA from Assay Genie uses a double-antibody, colorimetric format for the quantitative detection of Interleukin-9 (IL-9) in rabbit serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Performance characteristics include a sensitivity of 9.375 pg/mL and a working range of 15.625-1000 pg/mL. Linearity in serum (n=5) spans 89-103% at 1:2, 88-101% at 1:4, and 86-104% at 1:8; EDTA plasma (n=5) shows 88-101%, 82-99%, and 84-97% at the same dilutions; and heparin plasma (n=5) shows 90-100%, 85-100%, and 84-93%. Reported recovery averages are 94% for serum (range 86-100%), 99% for EDTA plasma (range 91-104%), and 97% for heparin plasma (range 91-102%).

The assay protocol involves equilibrating reagents and TMB substrate to room temperature, a 90-minute primary incubation at 37°C, 60-minute biotin-labeled detection antibody binding, 30-minute HRP-streptavidin binding, 10-20 minute TMB substrate development in the dark, and absorbance measurement at 450 nm after adding the stop solution. Sample preparation includes centrifuging serum at 1000 × g for 20 minutes, plasma at 1000 × g for 15 minutes at 2-8°C, and cell culture supernatant at 2500 rpm for 5 minutes.
