Description

Designed for the quantitative detection of rabbit interleukin-5, this Assay Genie sandwich ELISA employs a double-antibody format and a colorimetric read-out at 450 nm. The kit is reactive with rabbit samples and has been validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Reported sensitivity is 9.375 pg/mL with a measurable range of 15.625–1000 pg/mL. Reagents are supplied for storage at 2-8°C for up to 12 months.

The assay workflow involves equilibrating reagents to room temperature, loading standards, samples and blanks onto the pre-coated plate, and incubating at 37°C for 90 minutes. A biotin-labelled detection antibody is then added for a 60-minute incubation at 37°C, followed by HRP-Streptavidin (SABC) for 30 minutes, TMB substrate development in the dark for 10–20 minutes, and a stop reaction read at 450 nm. Linearity in serum, EDTA plasma and heparin plasma spans 83–105% across 1:2 to 1:8 dilutions (n = 5), and recovery averages 96% in serum, 97% in EDTA plasma and 95% in heparin plasma.

The kit lists IL-5 synonyms including IL5, EDF, TRF, E-CSF and ESGF, with the associated Uniprot identifier G1SL79. Sample preparation guidance is provided for serum (clot, 1000 × g, 20 minutes), plasma (anticoagulant tubes, 1000 × g for 15 minutes at 2–8°C), tissue homogenate, cell culture supernatant (2500 rpm for 5 minutes) and cell lysate. Included components comprise a dismountable pre-coated microplate, lyophilised standard, biotin-labelled antibody concentrate (100X), HRP-Streptavidin conjugate (SABC, 100X), TMB substrate, sample, antibody and SABC dilution buffers, stop solution, 25X wash buffer, plate sealer and a technical manual. Protocols are lot-specific; users are directed to the manual supplied with their kit.
