Description

From Assay Genie, this sandwich-format ELISA uses a double-antibody configuration to measure Interleukin-18 (IL-18), also known as Interferon-gamma-inducing factor, in rabbit samples. Reactivity is specific to rabbit, and the assay accepts serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Colorimetric detection is used, with a sensitivity of 9.375 pg/mL and a working range of 15.625-1000 pg/mL. Unopened kit components are stored at 2-8°C and remain stable for up to 12 months.

The protocol begins with equilibration of reagents and TMB substrate to room temperature, after which standards, samples, and a zero control are dispensed onto the pre-coated plate and incubated at 37°C for 90 minutes. A biotin-labelled detection antibody is then added for a 60-minute incubation at 37°C, followed by HRP-Streptavidin (SABC) for 30 minutes at 37°C. Colour development is performed with TMB substrate in the dark for 10-20 minutes, the reaction is stopped, and absorbance is read at 450 nm. Recommended preparation includes centrifugation of clotted blood at 1000 × g for 20 minutes for serum, and centrifugation of anticoagulant-collected blood at 1000 × g for 15 minutes at 2-8°C for plasma.

Assay linearity was demonstrated at 1:2, 1:4, and 1:8 dilution factors. For serum (n=5), recoveries ranged 85-100%, 87-99%, and 89-99% across the three dilutions. EDTA plasma (n=5) gave 82-101%, 87-96%, and 83-100%, while heparin plasma (n=5) gave 91-100%, 83-100%, and 82-98%. Overall spike-and-recovery across the same matrices produced the following ranges and averages: serum 85-101%, average 92%; EDTA plasma 93-98%, average 96%; heparin plasma 88-103%, average 93%.
