Description

Designed by Assay Genie for the precise measurement of HMGB1 in rabbit samples, this high-quality, sensitive ELISA kit supports serum, plasma, cell culture supernatant, cell lysis buffer or tissue lysis buffer, and other biological fluid samples. HMGB1 (High Mobility Group Box 1) is a key protein involved in inflammatory responses, tissue repair, and immune system regulation, and its dysregulation has been associated with diseases such as sepsis, arthritis, and cancer.

The kit uses a colorimetric Sandwich ELISA with a double-antibody format. It offers a sensitivity of 0.375 ng/mL and a working range of 0.625–40 ng/mL. Validated linearity values reach 89–100% (serum, n=5, 1:2 dilution), with recovery averaging 92% in serum, 96% in EDTA plasma, and 96% in heparin plasma. All kit components should be stored at 2–8°C, where they remain stable for 12 months.

The procedure follows a standard sandwich ELISA workflow: equilibrate reagents and the TMB substrate to room temperature, incubate the plate at 37°C for 90 minutes for primary antigen binding, add the biotin-labelled detection antibody and incubate at 37°C for 60 minutes, add HRP-Streptavidin (SABC) and incubate at 37°C for 30 minutes, develop with TMB substrate in the dark for 10–20 minutes, then add stop solution and read absorbance at 450 nm. Sample preparation protocols are supplied for serum, plasma, tissue homogenate, cell culture supernatant, and cell lysate.
