Description

Cardiac troponin I is a key cardiac marker used for the diagnosis and prognosis of acute coronary syndromes, myocardial infarction and other cardiovascular conditions. The Assay Genie sandwich ELISA kit provides a colorimetric, double-antibody assay for the quantitative measurement of rabbit cardiac troponin I (cTn-I/TNNI3) across a working range of 1.563–100 pg/mL, with a sensitivity of 0.938 pg/mL. Compatible sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit is stored at 2–8°C for 12 months and is supplied with all reagents required to run the protocol, including a pre-coated microplate, lyophilized standard, biotin-labelled detection antibody, HRP-streptavidin conjugate, TMB substrate, dilution and wash buffers, stop solution, plate sealers and a technical manual.

Validated performance is reported in serum, EDTA plasma and heparin plasma. Linearity across 1:2, 1:4 and 1:8 dilutions is 84–100% / 82–101% / 84–98% for serum, 85–101% / 87–104% / 86–102% for EDTA plasma and 91–102% / 83–99% / 82–95% for heparin plasma (n = 5 each). Spike-and-recovery ranges are 94–102% (mean 96%) for serum, 92–102% (mean 97%) for EDTA plasma and 93–105% (mean 99%) for heparin plasma. The assay uses a six-step protocol: equilibrate reagents and the TMB substrate to room temperature, then load standards, samples and a blank onto the pre-coated plate and incubate at 37°C for 90 minutes; add biotin-labelled detection antibody and incubate at 37°C for 60 minutes; add HRP-streptavidin (SABC) and incubate at 37°C for 30 minutes; develop with TMB substrate in the dark for 10–20 minutes; and finally add stop solution and read the absorbance at 450 nm.

Sample preparation guidance is provided for serum (allow blood to clot, centrifuge at 1000 × g for 20 minutes and collect the supernatant), plasma (collect in anticoagulant tubes, centrifuge at 1000 × g for 15 minutes at 2–8°C), tissue homogenate (homogenize in PBS with protease inhibitors and collect the supernatant), cell culture supernatant (centrifuge at 2500 rpm for 5 minutes) and cell lysate (lyse using a buffer with protease inhibitors and collect the protein supernatant).
