Description

A sandwich ELISA from Assay Genie for the quantitative measurement of cathepsin K in rabbit samples. Cathepsin K is a key enzyme in bone metabolism, responsible for collagen degradation in bone tissue, and its dysregulation has been linked to conditions such as osteoporosis and osteoarthritis, making it a valuable biomarker for research into bone remodelling and related disorders.

The assay uses a double-antibody sandwich format with colorimetric detection. Acceptable sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit reports a sensitivity of 0.938 pmol/L and a working range of 1.563–100 pmol/L (31.25–2000 pg/ml). Spiking recovery averages 95% for serum (range 88–102%), 98% for EDTA plasma (range 89–105%), and 93% for heparin plasma (range 86–105%), with linearity across 1:2, 1:4 and 1:8 dilutions of 86–103%, 83–95% and 80–94% for serum, 88–99%, 82–98% and 90–96% for EDTA plasma, and 86–96%, 83–100% and 89–99% for heparin plasma. All components are intended for storage at 2–8°C for 12 months.

The workflow involves equilibrating reagents to room temperature, loading standards and samples for a 90-minute primary incubation at 37°C, adding biotin-labelled detection antibody for 60 minutes, then HRP-streptavidin for 30 minutes, developing with TMB substrate for 10–20 minutes in the dark, and reading absorbance at 450 nm after adding stop solution.
