Designed for RNA-seq library construction from single cells and low-input RNA samples, this library preparation solution from QIAGEN enables researchers to investigate the full transcriptome with flexible input options. The workflow supports unbiased amplification and unbiased NGS library construction, allowing reliable investigation of the transcriptome from single cells, low input RNA samples, and small viral RNA genomes.

An important feature is the use of multiple-displacement amplification (MDA) to uniformly amplify cDNA alongside PCR-free NGS library construction. The MDA reaction uses a unique HiFi polymerase that minimizes the incorporation of amplification errors. The kit provides a time-saving, one-tube library preparation protocol that eliminates sample cleanup between steps, reducing starting material loss and cross-contamination risk. Co-optimization of MDA and NGS library construction processes enables a highly streamlined and efficient protocol that can be easily automated, producing NGS-ready libraries in one working day.

Intended for molecular biology applications, the kit is not intended for the diagnosis, prevention, or treatment of a disease. The workflow includes cell lysis and gDNA removal, reverse transcription using Quantiscript RT Enzyme Mix for 60 minutes, ligation of cDNAs for 30 minutes, and an isothermal cDNA amplification reaction proceeding for 120 minutes. Amplified cDNA can be stored long-term at –30 to –15°C. The enzyme kit and UDI Y-Adapter Kit should be stored at –30 to –15°C upon receipt, while the beads should be stored at 2–8°C and never frozen. The kit is compatible with Illumina NGS instruments.
