Description

From Assay Genie, this cell-based ELISA kit provides a sensitive and specific method for detecting and quantifying phosphorylation of PYK2 at Tyrosine 881 in cultured cells. The kit is intended for research into PYK2 signalling in areas such as cancer, inflammation, and neuronal function.

Detection is achieved through an indirect ELISA format in which PYK2 (Phospho-Tyr881) is captured by phospho-specific primary antibodies and visualised via HRP-conjugated secondary antibodies that catalyse a colourimetric reaction at 450 nm upon substrate addition. The assay is described as lysate-free and high-throughput, and is suitable for adherent cells. It can be used to measure relative levels of PYK2 phosphorylation as well as to screen the effects of treatments, inhibitors (such as siRNA or chemicals), or activators. Reactivity is reported for Human, Mouse, and Rat samples, with a stated dynamic range of > 5000 Cells.

Two normalisation approaches are supported: an included monoclonal antibody specific for human GAPDH serves as an internal positive control, and a Crystal Violet whole-cell staining step can be used to determine cell density so absorbance readings can be corrected for plating differences. The associated gene is PTK2B (Gene ID: 2185, UniProt ID: Q14289), encoding a nonreceptor tyrosine kinase of the Fak family that is involved in processes such as cell migration, proliferation, and differentiation.
