Description

This Cell-Based ELISA Kit from Assay Genie is designed for the qualitative measurement of PTP1B protein phosphorylation at serine 50 in cultured cells. The assay uses an indirect ELISA format with phospho-specific primary antibodies and HRP-conjugated secondary antibodies, delivering a lysate-free, high-throughput workflow that avoids the need for cell lysis. Detection is by a colorimetric reaction read at 450 nm, and the kit is reactive with human, mouse and rat samples. It is intended for use with adherent cells and has a stated dynamic range of > 5000 Cells.

Two normalisation methods are built into the workflow. A monoclonal anti-GAPDH antibody is included as an internal positive control to calibrate target absorbance values, while a Crystal Violet whole-cell staining step allows users to normalise readings to cell density and account for plating variation. The kit can be used to monitor changes in PTP1B phosphorylation following treatment with inhibitors such as siRNA or chemical compounds, as well as activators, supporting studies into cell signalling mechanisms and conditions including cancer, diabetes and inflammatory disorders.

PTP1B is a non-receptor protein tyrosine phosphatase encoded by the PTPN1 gene. It is reported to negatively regulate insulin signalling, modulate the unfolded protein response and influence processes such as cell growth, differentiation and cytoskeletal reorganisation, making it a target of interest in metabolic and oncological research.
