PTP1B Colorimetric Cell-Based ELISA Kit

From Assay Genie, this colorimetric cell-based ELISA kit enables qualitative measurement of protein tyrosine phosphatase 1B (PTP1B) levels directly in cultured cells. Designed as a lysate-free, high-throughput assay, it detects the relative expression profile of PTP1B in adherent cells and can be used to screen the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on PTP1B. The kit is reactive with human, mouse, and rat samples.

Detection uses an indirect ELISA format in which PTP1B is captured by PTP1B-specific primary antibodies, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody to catalyze a colorimetric reaction at 450 nm upon substrate addition. The assay has a dynamic range of > 5000 cells and is supplied in a 96-well microplate format, with a storage stability of 4°C for 6 months.

A monoclonal anti-GAPDH antibody is included as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining method may be used to determine cell density, allowing absorbance values to be normalised to cell amounts and plating differences to be adjusted.
